A 1:250 dilution of SPC-1139 was sufficient for detection of ErbB4 in 10 µg of mouse brain lysate by ECL immunoblot analysis using goat anti-rabbit IgG:HRP as the secondary antibody.
Cell membrane
Expressed at highest levels in brain, heart, kidney, in addition to skeletal muscle, parathyroid, cerebellum, pituitary, spleen, testis and breast. Lower levels in thymus, lung, salivary gland, and pancreas. Isoform JM-A CYT-1 and isoform JM-B CYT-1 are expressed in cerebellum, but only the isoform JM-B is expressed in the heart.
ErbB4 is an RTK involved in cell growth and differentiation. Ligands are neuregulins and not EGF. Donwregulation has been described in bladder cancer to correlated with oncogensis, but upregulation of ErbB4 is noted in lung cancer. Its mutation pattern in human tumours is very characteristic of a tumour suppressor protein.
